As a PhD student, my project focuses on native mass spectrometry of intact protein and protein complexes from micro-organisms with the intention of analyzing the proteins through direct MS from living colonies. A method will be developed to enable the detection of microbial intracellular proteins or membrane proteins; different technologies such as liquid surface extraction analysis (LESA) MS, electroporation technique and high-field asymmetric waveform ion mobility separation (FAIMS) will be potentially used, to allow the in-situ analysis of native protein from bacterial colonies. A further goal will be exploring the method as a tool for research of microbial biological process or diagnosis for the identification of microbes.
